For a rather classical story/poem:

Centered in your story must be the "fatal flaw". A misjudgement made by your character. This could be anything, for example your character chose the wrong lover, or hubris. This mistake haunts the character as it progresses in the story. In the end your character must face his fault, and overcome it. Or die of it.
In the classical tragedy the story must be sad and moralistic, should it have a proper working on the viewer/reader. A cleaning working that is. The spectator can relate to the fact that the problems in his/her life are worse than the problems in the characters life.
This is often achieved by everyone dying.
